CrtE is the gene which encodes geranylgeranyl diphosphate synthase, an essential enzyme in the MVA pathway in plants, through which paclitaxel is eventually synthesized. After GPP is converted into FPP, geranylgeranyl diphosphate synthase converts it into GGPP, an important precursor in the MVA pathway. Taxadiene synthase is the enzyme which makes the precursor to paclitaxel, taxadiene, from GGPP. It is naturally occurring, found in the Taxus genus1. We specify that this taxadiene synthase is cytosolic rather than from the MEP pathway in the chloroplast, which would require us to add export tags and make our engineering goal more difficult to reach.
